他達那非合成研究 |

摘 要:他達那非為口服磷酸二酯酶5(PDE 5)抑制劑,用于治療男性勃起功能障礙(MED)。筆者以D-色氨酸為原料,先甲酯化反應,生成的D-色氨酸甲酯鹽酸鹽在異丙醇中與胡椒醛發(fā)生Pictet-spengler反應,得到順式四氫咔啉化合物,然后再依次與氯乙酰氯和甲胺反應,總收率為56%,經(jīng)過(guò)篩選,冰醋酸作為重結晶溶劑進(jìn)行進(jìn)一步純化。 |

Study on the Synthesis of Tadalafil |

Abstract:Tadalafil is a new phosphodiesterase 5 inhibitor used for the treatment of male erectile dysfunction(MED). For the preparation of tadalafil,D-tryptophan was used as the starting material. It was converted to its methyl ester,then underwent Pictet-spengler reaction with heliotropin in isopropanol to give the cis-tetrahydrocarboline,which was further reacted with chloroacetyl chloride,followed by methylamine. Tadalafil was obtained in an overall yield of 56% and was recrystallized from acetic acid. |
