氨噻肟酸乙酯的合成研究 |

摘 要:以乙酰乙酸乙酯為原料,經(jīng)肟化、甲基化、鹵化及環(huán)合合成了(Z)-2-(2-氨基-4-噻唑)-2-甲氧基亞胺基乙酸乙酯。其中,肟化、甲基化無(wú)需分離提純,簡(jiǎn)化了操作步驟,鹵化過(guò)程用磺酰氯代替了傳統的鹵化試劑溴素,降低了反應的成本,并考察鹵化反應中各因素對收率的影響。采用紅外、核磁及液質(zhì)連用色譜等確定其結構,通過(guò)高效液相色譜測定該工藝制得的氨噻肟酸乙酯的純度可達到98%以上。產(chǎn)品的總收率達到46%(以乙酰乙酸乙酯計)。 |

Abstract:2-(2-Aminothiazol-4-yl)-(Z)-2-(methoryimion)acetate was synthesized from ethyl acetoacetate by oximation,methylation,halogenation and cyclic reaction in overall yield of 46%. The products from oximation and methylation were not necessary of separation and purification,therefore the process was simplified. Replacement of bromine with sulfuryl chloride as the halogenating reagent reduced the costs of the reaction. The influences of different factors on chlorination were investigated. The structure of the title compound was determined with FT-IR,1H NMR and LC-MS. |
