生物催化不對稱(chēng)還原制備(R)-2-羥基-4-苯基丁酸 |

摘 要:以克隆表達的乳酸脫氫酶(D-LDH)為催化劑來(lái)還原前手性化合物2-羰基-4-苯基丁酸(OPBA),制備重要的手性中間體(R)-2-羥基-4-苯基丁酸(R-HPBA)。通過(guò)考察影響反應的各因素,確定了反應pH、初始底物濃度,酶加量及NAD+加量等參數。最后,以分批補料的方式進(jìn)行產(chǎn)物制備,4 h內即可完全轉化100 mmol/L底物。經(jīng)驗證,所得產(chǎn)物的構型完全正確,且e.e值>99.9%。 |

Preparation of(R)-2-Hydroxy-4-phenyl-butyric Acid by Biocatalytic Asymmetric Reduction |

Abstract:The important chiral intermediate(R)-2-hydroxy-4-phenyl butyric acid(R-HPBA)was prepared from prochiral compound,using lactate dehydrogenase(D-LDH)obtained by cloning and expression as the biocatalyst. The affecting factors such as pH,initial substrate concentration,enzyme loading,and NAD+ amount were investigated. Finally,the product was prepared from 100 mmol/L substrate in 4 h by a fed-batch process. The configuration of the target product obtained therein was confirmed,and the e.e > 99.9%. |
