蛋白酶體抑制劑的研究進(jìn)展 |

| 摘 要：隨著(zhù)近期美國食品和藥物管理局（FDA）批準將Carfilzomib用于難治性多發(fā)性骨髓瘤患者的治療，20 S蛋白酶體已經(jīng)成為一種越來(lái)越引人關(guān)注的腫瘤治療靶標。目前，已經(jīng)開(kāi)發(fā)出了一系列低納摩爾濃度的合成和天然20 S蛋白酶體抑制劑，作為抗癌和抗炎的藥物進(jìn)入各期臨床實(shí)驗。此外，蛋白酶抑制劑繼續作為有價(jià)值的研究工具，通過(guò)闡明蛋白質(zhì)在泛素-蛋白酶體降解途徑這一重要的生物學(xué)過(guò)程，在細胞生物學(xué)中發(fā)揮作用。 |

| The Research Progress of Proteasome Inhibitors |

| Abstract：With the approval of carfilzomib for the treatment of refractory multiple myeloma patients by FDA，the proteasome has been become one of the most attractive targets for treatment of cancer. So far，a vast array of synthetic and natural compounds have been developed with low nanomolar activity as the 20S proteasome inhibitors and entered pre-clinic or clinical trials as anti-cancer and anti-inflammatory agents. In addition，the protease inhibitors are used as the research tool to clarify the process of protein degradation in the ubiquitin - proteasome pathway. This review will highlight the latest progress on the research of proteasome inhibitor. |
