四對硝基苯基卟啉和四對氨基苯基卟啉的合成

摘 要:分別采用苯基卟啉硝化法(兩步法)和對硝基苯甲醛直接合成法合成了四對硝基苯基卟啉(TNPP)。苯基卟啉硝化法首先采用Alder方法合成四苯基卟啉,然后以發(fā)煙硝酸為硝化試劑,將苯環(huán)對位硝化,反應產(chǎn)物是一硝化、二硝化、三硝化和四硝化產(chǎn)物的混合物,難分離,而且此路線(xiàn)腐蝕性強,操作困難,不宜大規模制備。對硝基苯甲醛直接合成法,以丙酸做反應溶劑,使反應活性降低,從而避免了更多的副反應,反應結束后,直接過(guò)濾即可得到四對硝基苯基卟啉,收率為22%。再將TNPP用二水合氯化亞錫在鹽酸中還原得到四對氨基苯基卟啉(TAPP),還原收率為60%。IR、1H NMR、元素分析等證實(shí)產(chǎn)物結構。紫外可見(jiàn)吸收光譜和熒光光譜顯示相對于四苯基卟啉,吸電子的硝基和給電子的氨基均使其Q帶和B帶的吸收位置紅移,并使其發(fā)光位置也明顯紅移。

Synthesis of Tetra(4-aminophenyl)porphyrin and Tetra(4-nitrophenyl)porphyrin

Abstract:Porphyrins play an important role in materials science,special function of analytical reagents,electrochemicals and life science. Preparations of tetra(4-nitrophenyl)porphyrin(TNPP)starting from benzaldehyde(method 1)and from nitrobenzaldehyde(method 2)were investigated. In method 1,the tetraphenyl porphyrin was synthesized by the reaction of benzaldehyde with pyrrole and then nitrated by concentrated sulfuric acid and fuming nitric acid. The nitration products include mono-,di-,tri- and tetra-nitrophenyl porphyrin,which are uneasy to be separated. Moreover,strong corrosive acids make it difficult for large scale production. In method 2 when the reaction of nitrobenzaldehyde with pyrrole was conducted in propionic acid,many side reactions could be avoided,thus making the work-up simple. Reduction of TNPP with stannous chloride hydrate in hydrochloric acid resulted in tetra(4-aminophenyl)porphyrin in 60% yield. The chemical structures were characterized by IR,1H NMR and elemental analysis.
