特拉匹韋中間體的合成 |

| 摘 要：L-環(huán)己基甘氨酸甲酯與吡嗪-2-甲酰氯進(jìn)行酰胺化反應后水解成（2S）-2-環(huán)己基-N-（2-吡嗪基羰基）甘氨酸，再與L-叔亮氨酸甲酯縮合，最后經(jīng)水解得到特拉匹韋關(guān)鍵中間體（2S）-2-環(huán)己基-N-（2-吡嗪基羰基）甘氨酰-3-甲基-L-纈氨酸，總收率68.3%。 |

| Abstract：（S）-2-（（S）-2- Cyclohexyl -2-［（pyrazine-2-carbonyl）-amino］-acetylamino）-3，3-dimethyl-butyric acid，the key intermediate for Telaprevir，was synthesized from L-cyclohexyl glycine methyl ester by reaction with 2-pyrazinecarbonyl chloride，followed by hydrolysis，condensation with L-tert-leucine methyl ester，and hydrolysis of the ester. The
overall yield of the target product was 68.3%. |
