新型PBK/TOPK抑制劑OTS514的合成 |

摘 要:以3-溴噻吩-2-甲酸為起始原料,經(jīng)酰氯、鹵代、酰胺化、氨基保護、分子內環(huán)化、溴代得中間體9-溴-8-甲氧基-6-甲基噻吩并[2,3-c]喹啉-4(5H)-酮;以(R)-2-苯基丙胺鹽酸鹽為原料,經(jīng)中和游離、;Wo、溴代、水解去保護、氨基Boc保護、親核取代得到(R)-(2-(4-(4,4,5,5-四甲基-1,3,2-二惡硼烷-2-基)苯基)丙基)氨基甲酸叔丁酯;2個(gè)中間體經(jīng)交叉偶聯(lián)反應、脫保護得OTS514,純度99.1%,總收率為24.7%,并采用1H NMR、13C NMR和MS進(jìn)行其結構確證。該合成路線(xiàn)反應條件溫和,適合于實(shí)驗室研究的小規模制備。 |

Synthesis of New PBK/TOPK Inhibitor OTS514 |

Abstract:9-Bromo-8-methoxy-6-methylthieno[2,3-c]quinolin-4(5H)-one, a key intermediate of OTS514, was synthesized from 3-bromothiophere-2-carboxylic acid via halogenation, acylation, amino protection, cyclization of thiophene [2,3-c] quinolone and bromo-substitution. Another crucial intermediate tert-butyl(R)-(2-(4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)phenyl)propyl)carbamate was obtained from (R)-2-phenylpropylamine hydrochloride via neutralization, acylation, bromo-substitution, hydrolysis, amino protection and nucleophilic substitution. Then these two intermediates were subjected to cross-coupling reaction and deprotection to afford the OTS514 with an overall yield of 24.7% and a purity of 99.1%. And the final production was characterized using 1H NMR, 13C NMR and MS. This process was of mild reaction condition and very suitable for lab-scale production. |
