他達那非關(guān)鍵中間體（1R，3R）-1-（3，4-亞甲基二氧苯基）-2，3，4，9-四氫-1H-吡啶并［3，4-b］吲哚-3-甲酸甲酯鹽酸鹽的合成 |

| 摘 要：以D-色氨酸為原料，通過(guò)甲酯化和Pictet-spengler 2步反應，合成了他達那非關(guān)鍵中間體（1R，3R）-1-（3，4-亞甲基二氧苯基）-2，3，4，9-四氫-1H-吡啶并［3，4-b］吲哚-3-甲酸甲酯鹽酸鹽，2步反應總收率為87.4%。其結構經(jīng)核磁共振氫譜（1H NMR）和質(zhì)譜（ESI-MS）確證�？疾炝巳軇�、物料比、溫度和反應時(shí)間4個(gè)因素對Pictet-spengle反應的影響。結果表明，在反應溶劑為異丙醇、物料比n（胡椒醛）∶n（D-色氨酸甲酯鹽酸鹽）= 1.3∶1.0、90℃反應13 h的優(yōu)化條件下，目標產(chǎn)物的收率達92.5%。 |

| Abstract：(1R,3R)-Methyl 1-(benzo[d][1,3]dioxol-5-yl)-2,3,4,9-tetrahydro-1H-pyrido[3,4-b]indole-3-carboxylate hydrochloride, the key intermediate of Tadalafil, was synthesized from D-tryptophan through a two-step reaction including esterification and Pictet-spengler reaction with a total yield of 87.4%. The structure of the product and intermediate were confirmed by 1H NMR and ESI-MS. The optimal reaction conditions of Pictet-spengler reaction were obtained as followings: isopropanol was used as solvent; the optimal molar ratio of piperonyl aldehyde and D-tryptophan methyl ester hydrochloride was 1.3; the mixture was reacted at 90℃ for 14 h. The product (Tadalafil intermediate) was obtained with a yield of 92.5% under optimal reaction condition. |
