光譜法研究2-乙基-3-甲基-吡嗪與牛血清白蛋白的相互作用 |

摘 要:利用熒光和紫外光譜法在模擬生理條件下,研究了牛血清白蛋白(BSA)和2-乙基-3-甲基-吡嗪(NEMP)的相互作用。應用Stern-Volmer方程、Lineweaver-Burk雙倒數方程及F?觟rster非輻射能量轉移理論,計算獲得3個(gè)不同溫度下(288、298、308 K)兩者的猝滅常數(KSV)、結合常數(Kq)、結合距離(r)和熱力學(xué)參數(ΔH,ΔG和ΔS)等。結果顯示:在不同溫度下,BSA 和NEMP均能相互結合生成新的復合物,且屬于靜態(tài)猝滅;結合距離小于7 nm;同步熒光顯示NEMP對BSA的構象產(chǎn)生影響,主要是對其中酪氨酸殘基有顯著(zhù)的影響;而在紫外光譜中BSA的最大吸收峰向較短波長(cháng)偏移,BSA與NEMP之間形成了基態(tài)配合物。 |

Spectroscopic Study on the Interaction of a Novel 2-Ethyl-3-methylpyrazine(NEMP) and Bovine Serum Albumin |

Abstract:The interaction mechanism of bovine serum albumin (BSA) with 2-ethyl-3-methylpyrazine (NEMP) at three different temperatures was studied in Tris-HCl buffer solution at pH7.43. Refering to F?觟rster non-radioactive energy transfer theory, the Stern-Volmer quenching constant(KSV), the bimolecular quenching rate constant (Kq), binding distance (r) and the thermodynamic parameters (ΔH, ΔG and ΔS) between NEMP and BSA were measured. The results showed that BSA and NENP could form new complexes at three temperatures with static quenching mechanism. The binding distance was less than 7 nm. Synchronous fluorescence spectra showed that NEMP had an effect on the conformation of BSA, mainly on the Tyr residues. The maximum absorption peak of BSA in the ultraviolet absorption spectrum was shifted, and the ground state complex was formed between BSA and NEMP. |
