氮雜環(huán)醇制備方法及其藥物合成中的應用進(jìn)展 |

摘 要:氮雜環(huán)醇(Azacyclonol)屬于含有氮雜環(huán)的二苯甲醇行生物, 具有良好的反應活性, 廣泛應用于有
機合成、 藥物及特種材料制備等領(lǐng)域。 綜述了氮雜環(huán)醇的合成, 依據起始物料比較了以 4-咄唗甲酸乙趴、4-派唗甲酸、 N-乙;蓡t-4-;、 4-氛基咄唗為原料制備氮雜環(huán)醇的 4 條合成路線(xiàn)。
其中, 以 4- 派唗甲酸路線(xiàn)較優(yōu), 具備工時(shí)短、 能耗低、 物料價(jià)廉易得、 操作簡(jiǎn)便之特點(diǎn), 宜于工業(yè)化大生產(chǎn)。 同時(shí)歸納了氮雜環(huán)醇在藥物合成中的應用并展望了其發(fā)展趨勢。
為探究更經(jīng)濟、 高效的氮雜環(huán)醇合成方法及下游醫藥產(chǎn)品開(kāi)發(fā)提供借鑒意義。

Abstract:Azacyclonol is a derivative compound of piperidine benzhydrol with high reactivity and
widely used in
research fields such as organic synthesis, pharmaceutical preparation, and special materials. In
this paper, the synthetic routes of azacyclonol in recent years were reviewed. Based on different
starting materials including ethyl 4 -picolinate, 4 -piperidine carboxylic acid, N
-acetylpiperidine -4 -acid chloride, and 4 -cyano, four synthetic routes for the preparation of
azacyclonol were analyzed including their advantages and disadvantages. Among these four synthesis
methods, the 4 -piperidine carboxylic acid route was the best one, which had the characteristics
including short working hours, low energy consumption, low cost and easy availability of raw
materials, and simple operation. It had advantages in large-scale industrial production. At the
same time, azacyclonol in drug synthesis was summarized, and its development trend was prospected.
It provided reference for exploring economical and efficient synthesis methods of azacyclonol and
the development of downstream products.
