HPLC-RID法測定磷酸奧司他韋干混懸劑中山梨醇的含量 |

摘 要:磷酸奧司他韋是目前臨床上治療禽流感、甲型H1N1流感病毒和乙型流感病毒最有效的藥物之一。采用高效液相色譜-示差折光檢測法測定磷酸奧司他韋干混懸劑中關(guān)鍵輔料山梨醇的含量。優(yōu)選色譜柱Sepax Technologies carbomix Ca-NP 10∶8%(7.8 mm×300 mm,10 μm),流動(dòng)相為超純水,流速0.5 mL/min,色譜柱柱溫80 ℃,檢測器溫度55 ℃,進(jìn)樣量20 μL。結果顯示:磷酸奧司他韋干混懸劑中除山梨醇之外的其余混合輔料及原料藥均不干擾山梨醇的檢測;山梨醇在一定濃度范圍內呈良好線(xiàn)性關(guān)系;重復測試6份樣品,相對標準偏差0.53%,回收率99.36%~100.33%,RSD為0.30%。所建立的檢測方法適用于磷酸奧司他韋干混懸劑中山梨醇含量的測定。 |

Determination of Sorbitol in Oseltamivir Phosphate Suspension by HPLC-RID |

Abstract:Oseltamivir phosphate is currently one of the most effective drugs for avian influenza, influenza A (H1N1) virus and influenza B virus. The content of sorbitol in oseltamivir phosphate suspension was determined by HPLC-RID on the column Sepax Technologies carbomix Ca-NP 10:8% (7.8 mm×300 mm, 10 μm). The separation conditions included: the mobile phase is water; the flow rate was 0.5 mL/min; the column temperature was 80 ℃; the detector temperature was 55 ℃; and the injection volume was 20 μL. The test results showed that the other mixed excipients and API in oseltamivir phosphate suspension other than sorbitol do not interfere with the detection of sorbitol. Sorbitol showed a good linear relationship. Six samples were tested repeatedly, with a relative standard deviation of 0.53%, the recovery rate is 99.36%~100.33%, RSD is 0.30%. The established method is suitable for the determination of sorbitol in oseltamivir phosphate suspension. |
