山豆根提取物質(zhì)量標準研究

摘 要:建立山豆根提取物的質(zhì)量標準。選取乙醇濃度、料液比、回流提取時(shí)間及提取次數為影響因素,采用正交實(shí)驗法優(yōu)化山豆根提取物提取工藝,并以HPLC法測定山豆根提取物中苦參堿和氧化苦參堿的含量。優(yōu)化提取工藝為:乙醇濃度60%、料液比1∶20(g∶mL)、回流時(shí)間2 h、提取次數3次。HPLC含量測定結果為:苦參堿及氧化苦參堿在選定范圍內線(xiàn)性關(guān)系良好,加樣回收率RSD不高于2%。提取工藝穩定可行,且建立的HPLC含量測定方法簡(jiǎn)便、快捷。

Study on the Quality Standard of Sophora Tonkinensis Gagnep Extract

Abstract:The quality standard of the extract from the Sophora tonkinensis Gagnep was established. Ethanol concentration, solid-liquid ratio, reflux time and extraction times were selected as the influencing factors. Orthogonal experiment was used to select the extraction technology of the Sophora tonkinensis Gagnep, and the content of matrine and oxymatrine in the Sophora tonkinensis Gagnep was determined by HPLC. Orthogonal test results showed that the optimal extraction process was as followings: ethanol concentration (was 60%), solid-liquid ratio was 1∶20 (g∶mL), reflux time was 2.0 h, extraction times were 3 times. HPLC results showed that matrine and oxymatrine had good linear relationship in the selected range, and the recovery RSD was not higher than 2%. The extraction process is stable and feasible, and the established HPLC method is simple and fast.
