用畢氏酵母表達蛇島蝮蛇毒類(lèi)凝血酶發(fā)酵條件的優(yōu)化 |

| 摘 要：為了提高大連蛇島蝮蛇毒類(lèi)凝血酶的表達量 ,利用 5 0 0ml搖瓶培養體系研究了在不同培養基、pH值、發(fā)酵時(shí)間等培養條件下分泌蛋白的濃度及活性 ,使發(fā)酵條件得到優(yōu)化 :豐富培養基比營(yíng)養缺陷型培養基更利于目的蛋白的分泌表達 ,發(fā)酵優(yōu)惠條件 pH值為 6 0 ,時(shí)間為 36h ,發(fā)酵液上清能檢測到靶酶的酰胺水解活性。類(lèi)凝血酶的表達量約為 4mg/L。 |

| Abstract：To increase the expression yield of Gloshedobin by Pichia pastrois,the recombinant thrombin-like enzyme was expressed ith 500ml shaking flask.By studying the concentration and activity of secreted protein under different conditions:components of medium,pH,and the time of fermentation,the optimal fermentation conditions were obtained:rich medium is more beneficial for secretion expression than minimal medium,the optimal pH 6.0, and the optimal fermentation time 36 hours.The amidolytic actvity can be detected... |
