高效液相色譜法測定蛋黃卵磷脂中膽固醇的含量 |

| 摘 要：應用高效液相色譜 蒸發(fā)光散射檢測器測定蛋黃卵磷脂中膽固醇含量。采用Hy persilDOSC18(150mm×4.6mm,5μm)色譜柱,以乙腈∶甲醇∶異丙醇=10∶80∶10(v/v)為流動(dòng)相,流速為1ml/min,柱溫為25℃,蒸發(fā)光散射檢測器的空氣流速為2L/min,漂移管溫度為85℃。膽固醇進(jìn)樣量在0.22～1.83μg的范圍內與峰面積的對數呈良好的線(xiàn)性關(guān)系。相關(guān)系數為0.9998,回收率為99.59%。方法操作簡(jiǎn)便,精密度好,結果準確可靠。樣品的測定結果為0.42%。 |

| Determination of Cholesterin in Leicithin from Egg with HPLC-ELSD |

| Abstract：The Cholesterin in Leicithin from egg was analysized with HPLC ELSD, The conditions are as follow: using hypersil ODS C 18 Column(150?mm×4.6?mm, 5?μm), CH 3CN∶CH 3OH∶(CH 3) 2CHOH=10∶80∶10 as mobile phase, the flow rat 1?ml·min -1 , the temperature of column 25?℃, the gas flow rate 2?L·min -1 , the tube temp 85?℃. The calibration curve is linear in the range of 0.22～1.83?μg for cholesterin(r=0.9998). The average recovery(n=5) is 99.53%. The method is rapid, accurate, and reliable. |
